Sudan Black B Stain (SBB)

Intended Use:

This kit is for staining bone marrow and blood cellsmear.

 

Principle:

Sudan Black B is a fat-soluble stain used for staining triglycerides and lipids. In cytopalsm, Sudan Black B can stain granules dark brown.

Methods:

1. SBB stain is designed for dip method.

2. Fix dried bone marrow and blood smear in Fixative for 10~20 seconds. Rinse with distilled water. Air dry the

    smear.

3. Dip the smear into Sudan Black B solution and incubate at 37℃  for 20~30 minutes. Rinse with distilled

    water; air dry or drywith filter paper.

4. Counterstain with Wright Stain. (Stain the smear with Wright Stain and phosphate buffer solution (1:2) for

    5~10 minutes.Rinse withdistilled water. Dry the smear for microscopic examination.

 

Expected Results:

Dark brown granules found in cytoplasm means positive.
